We've changed every chemical we are working with in our 212aa PCR efforts. 3 weeks on, still no product. Having read Tavi last night, I came into class this morning with fresh ideas regarding denaturing, annealing and extension time/temperatures for both the T4 and Agro protocols. Ran some of these ideas out with others on both TAE and SB gels - nothing. Thoughts moving forward... Tags: J. Darland
