I don’t know why I didn’t think of this earlier. Sometimes troubleshooting is a round and round process. Suddenly you stare at something you’ve seen 1000 times and all of the sudden it looks different.
Josh did a great experiment yesterday. He ran a bunch of PCRs, some were done with the same buffers and reagents we have used in the past, some were done with a mastermix kit from Promega. (In earlier blogs we have called this ‘Dr. Obermiller’s Secret Sauce’.)

In the lab today I talked with one of the students as he looked at the results of his most recent PCR. On one gel he had 4 or 5 rxns showing the desired product 1464 base pairs.
