Its about 3:10 In the afternoon, and im sitting in the lab still. After a busy morning of doing all these little itty bitty (not a waste of time) lab exercises for BIO220, at 11:30 I could finally sit down and get some real work done.

I wish I saw this earlier

I don’t know why I didn’t think of this earlier. Sometimes troubleshooting is a round and round process. Suddenly you stare at something you’ve seen 1000 times and all of the sudden it looks different.

 

Josh did a great experiment yesterday. He ran a bunch of PCRs, some were done with the same buffers and reagents we have used in the past, some were done with a mastermix kit from Promega. (In earlier blogs we have called this ‘Dr. Obermiller’s Secret Sauce’.)

In the lab today I talked with one of the students as he looked  at the results of his most recent PCR.  On one gel he had 4 or 5 rxns showing the desired product 1464 base pairs.

Science... is science.

Nothing worthwhile is going to be done correctly on the first try. Walt Disney was fired by a newspaper editor because "...he lacked creativity...", and Thomas Edison attempted to create the light bulb 4,000 times before he finally succeeded! Abraham Lincoln lost 6 presidential elections before he was finally elected president, and when he did finally win the office of president, it was the only election he ever won!

I surprised to hear that mr. Darland. I'm am somewhat grateful that i took a day off from PCR. I was really counting on the thermopol buffer solving the problems the class has been having.
