proC and trp B gene sequences and primers
2009.11.03 09:46:14

Scholars,

Visit the Documents section in the left hand menu and click on 212AA.  In this folder are the sequences for and primers used to amplify the genes proC and trpB.

And They're Off!
2009.11.03 08:36:03

And They’re Off!

The students of BIO 212 AA have begun their cloning projects! Each student will PCR and subsequently clone the gene for T4 DNA Ligase as well as a biosynthetic gene from Agrobacterium radiobacter k84 into the plasmid vector pUC19. This semester we are looking at 2 A. radiobacter genes involved in proline (proC) and tryptophan (trpB) biosynthesis.

7 AM Mesa Community College Part II
2008.11.21 21:59:20

It's 7 AM on a Friday.  The campus is a little more desolate on Fridays, but half the 212AA class is waiting for me at the door to the laboratory.

I wish I saw this earlier
2008.11.17 06:00:04

I don’t know why I didn’t think of this earlier. Sometimes troubleshooting is a round and round process. Suddenly you stare at something you’ve seen 1000 times and all of the sudden it looks different.

Josh did a great experiment yesterday. He ran a bunch of PCRs, some were done with the same buffers and reagents we have used in the past, some were done with a mastermix kit from Promega. (In earlier blogs we have called this ‘Dr. Obermiller’s Secret Sauce’.)

Science, you gotta love it.  99.9% experiments don't produce satisfaction.

Sunday Morning Troubleshooting
2008.11.10 03:03:52

Sunday Morning, Mesa Community College-

The BIO 212AA students have been having trouble with their lab experiments. Currently we are trying to PCR the DNA Ligase gene from Bacteriophage T4. A couple of students have been able to make the desired product, but overall the class has been stymied. Even the students who have had success were not able to repeat their results.

7:00 AM, Mesa Community College (Part I)
2008.11.06 10:35:41

One of my favorite times to be on campus is in the morning, at 7:00 AM specifically. The campus is not so crowded, but the students that are present possess such vibrancy, they walk with a sense of determination and purpose.
