I don’t know why I didn’t think of this earlier. Sometimes troubleshooting is a round and round process. Suddenly you stare at something you’ve seen 1000 times and all of the sudden it looks different.
Josh did a great experiment yesterday. He ran a bunch of PCRs, some were done with the same buffers and reagents we have used in the past, some were done with a mastermix kit from Promega. (In earlier blogs we have called this ‘Dr. Obermiller’s Secret Sauce’.)

Greetings Biotech Thunderbirds, My name is Hal Hawkington, I've had several careers in my life which has made me the person that I am. For many years I practiced plastic surgery in Beverly Hills and Palm Springs, California. Interest in the healing process led me to study stem cells and tissue differentiation. Sunday Morning, Mesa Community College- The BIO 212AA students have been having trouble with their lab experiments. Currently we are trying to PCR the DNA Ligase gene from Bacteriophage T4. A couple of students have been able to make the desired product, but overall the class has been stymied. Even the students who have had success were not able to repeat their results. One of my favorite times to be on campus is in the morning, at 7:00 AM specifically. The campus is not so crowded, but the students that are present possess such vibrancy, they walk with a sense of determination and purpose.
